The school provides a bilingual education in Spanish and English, with students achieving fluency by the end of the Primary level. The curriculum spans Initial (ages 2-5), Primary (years 1-6), and Secondary (years 1-6) levels. In the Secondary stage, students prepare for Cambridge international examinations and study Portuguese as a third language. The academic programme is supported by digital tools, including multimedia rooms, computer labs, and a virtual learning environment.

Students in the Secondary level prepare for international Cambridge examinations. Additionally, the curriculum includes a third language, Portuguese, to prepare students for future professional challenges.

The school implements a Social Skills Programme designed to help students build lasting relationships, work productively and empathetically in groups, and adapt to social changes. This programme is integrated into daily academic life, focusing on cooperation, collaboration, and the valuing of diverse perspectives.

The admissions process requires families to complete an online form, which is a prerequisite for scheduling an interview. The process involves separate interviews with the admissions office and the relevant level director, followed by an evaluation or "play hour" for applicants to Kinder (ages 4-5), Primary, and Secondary levels. Results are typically communicated via email approximately 10 days after the evaluation.

St. Luke's College operates across three campuses: Olivos, Nordelta, and Haras Santa María. The Olivos campus features a computer lab, science lab, library, dining hall, auditorium, and specialized rooms for music, art, and audiovisual production, including a radio studio. The Nordelta campus, situated on a 2-hectare site, includes a computer lab, library, science lab, and sports field, while the 4-hectare Haras Santa María campus offers a multimedia room, art room, and sports fields for rugby and hockey.

The school offers a range of co-curricular activities including physical education, performing and visual arts, and a learning and service programme. Students are also organized into "Houses" to foster community and collaboration. Additionally, the school maintains a sports programme that utilizes both on-site fields and external facilities, such as the Olivos Rugby Club.
